Reconstitution of myeloid cells such as Granulocytes, monocytes, macrophages, dendritic and natural killer (NK) cells in immunodeficient mice is commonly recognized as challenging, primarily due to the absence of critical human cytokines. This often leads to relatively low levels of myeloid cells compared to T- and B-cell populations.​

Myeloid cell population is one of the key immune cells in immune system and play crucial roles in host defence against pathogens. They also have significant contributions to the pathogenesis of various diseases, including cancer, autoimmune disorders and infectious diseases. Therefore, myeloid-enhanced humanized mice provide a more relevant and accurate model for studying these diseases and developing new treatments.​

Improving the differentiation of T-cells, granulocytes, monocytes, macrophages, and eosinophils in humanized mice typically involves the use of the GM-CSF and IL-3 cytokine combination. However, introducing these cytokines using the transgenic method can be complex and expensive.

At our laboratory, we have developed a flexible, transient and cost-effective method to substantially enhance the myeloid cell population of CD34+ humanized mice. Our method increases the proportion of myeloid cells from 1-3% to 10-20% relative to hCD45+ cells. This enhanced population of myeloid cells makes these humanized mice ideal for studying immunotherapies targeting T-cells and monocytes and their functions in various disease settings.
